Bioactivity

名称	Epirubicin hydrochloride
描述	Epirubicin hydrochloride (Pharmorubicin) is an adriamycin derivative, a topoisomerase (Topo) inhibitor, and a Forkhead box protein p3 (Foxp3) inhibitor. Epirubicin hydrochloride has antitumor activity.
细胞实验	Hep G2 cells (500 cells/well, monolayer) are plated in a 96-well plate. The next day the cells are treated with Epirubicin in the medium. At the end of the incubation periods, 15% volume of MTT dye solution is added. After 1 hr of incubation at 37℃, an equal volume of solubilization/stop solution (dimethylsul-foxide) is added to each well for an additional 1 hr incubation. The absorbance of the reaction solution at 570 nm is recorded.(Only for Reference)
体外活性	方法: 神经胶质瘤细胞 U-87 和神经元原代培养物用 Epirubicin hydrochloride (0.5-100 µM) 处理 48 h，使用 MTT assay 检测细胞活力。结果: Epirubicin 以浓度依赖性方式显著降低大鼠 U-87 细胞的活力，IC50 为 6.3 µM。与 U-87 神经胶质瘤细胞相反，大鼠正常神经元细胞对 Epirubicin 具有耐药性。[1] 方法: 人乳腺癌细胞 MDA-MB-231 和 SK-BR-3 用 Epirubicin hydrochloride (2-3 µM) 处理 48 h，使用 Flow cytometry 检测细胞凋亡。结果: Epirubicin 在 G2/M 阻滞中显著增加了 sub-G 细胞。[2]
体内活性	方法: 为检测体内抗肿瘤活性，将 Epirubicin hydrochloride (7 mg/kg) 静脉注射给携带 MDA-MB-231 异种移植物的 BALB/c nu/nu 裸鼠，给药三次，间隔四天。结果: Epirubicin 将肿瘤生长减少了一半，同时使体重减轻了 10% 以上。Epirubicin 在 7 mg/kg 时的 IR 值为 53.3%。[2]
