Bioactivity

名称	Geniposide
描述	Geniposide is an iridoid glycoside with a variety of biological activities including neuroprotective, anti-diabetic, antiproliferative, and antioxidative activity. Geniposide has been shown to regulate Nrf2 translocation.
细胞实验	The third passages of brain microvascular endothelial cells (BMECs) are used for the experiment. The BMECs are divided into four groups: (1)normal control group: the normal cultured BMECs without treatment; (2)OGD group: the BMECs injured by OGD according to the above method; (3) geniposide group: the OGD-injured BMECs treated with 33.2 μg/mL geniposide for 6 h; (4)PTX group: the OGD-injured BMECs administrated with 100 ng/mL PTX. PTX, known as an inhibitor of Gi-coupled receptor is used to assess the activation of P2Y14 receptor induced by OGD in this experiment[2].
体外活性	Geniposide在多种生理活动中展现效果，包括抗血栓、抗炎、抗糖尿病、抗动脉粥样硬化、抗抑郁、改善阿尔茨海默病（AD）、降低高血压、毒理学性质及不良反应等[1]。在OGD诱导的脑微血管内皮细胞中，Geniposide显著降低了IL-8、IL-1β和MCP-1的产生，显著下调P2Y14受体的表达，并抑制RAF-1、MEK1/2、ERK1/2的磷酸化[2]。
体内活性	Geniposide（200及400 mg/kg）以剂量依赖性显著降低糖尿病小鼠的血糖、胰岛素和TG水平。该化合物还在mRNA及免疫活性蛋白水平和酶活性上降低GP与G6Pase的表达[3]。Geniposide（20.0、40.0或80 mg/kg）显著逆转因过量饮酒引起的血清ALT/AST和肝脏LPO水平的异常升高。Geniposide通过上调血红素加氧酶-1（HO-1）的表达，减轻3-吗啉基肼氯化物（SIN-1）在原代培养的海马神经元中诱导的细胞凋亡[4]。Geniposide抑制体内光化学诱导的血栓形成模型。Geniposide通过干扰IκB降解极为有效地抑制NF-κB[1]。
