Camptothecin is a quinoline alkaloid isolated from the Chinese endemic tree Camptotheca acuminata, and is the first natural topoisomerase I inhibitor discovered and confirmed to possess potent anti-tumor activity. It works by specifically inhibiting DNA topoisomerase I, interfering with DNA replication and transcription processes, leading to DNA damage and tumor cell apoptosis. Due to its unique pentacyclic structure and significant in vitro and in vivo anti-tumor activity, camptothecin is considered one of the most important natural anti-tumor drug lead compounds of the 20th century. Although it has poor water solubility and high toxicity itself, derivatives such as irinotecan, topotecan, and belotecan, optimized based on its parent nucleus structure, have become first-line clinical anti-tumor drugs. High-purity camptothecin and its key intermediates are indispensable core raw materials for modern anti-tumor new drug R&D, structural modification, mechanism of action research, and quality control.
