工艺路线总览:
上禾生物大蒜素/提取物依目标活性与稳定性分四条主线:
酶灭活固定蒜氨酸型(Alliin Type,人用口服/医药)——鲜蒜低温粉碎 → 微波/热灭酶(60~80℃短时或微波处理防Allicin瞬生降解) → 低温水/醇提 → 阳离子交换树脂/大孔树脂纯化 → 低温浓缩/冷冻干燥 → 得高蒜氨酸(Alliin 1%~98% HPLC)稳定无臭粉末;
保留酶活性型(Active Enzyme Type,食品/饲料替抗)——鲜蒜≤15℃低温粉碎 → 不加热不调pH → 立即冷冻干燥(-40℃升华)或≤35℃低温风干 → 粉碎包装,干燥状态下酶底物隔离,复水后蒜氨酸酶催化产Allicin;
稳定化大蒜素/微囊型(Stabilized Allicin / Microencapsulated)——酶解粗提 → β-环糊精包合/变性淀粉(N-LOK)纳米射流雾化微囊化/肠溶衣 → 低温干燥,锁住Allicin活性与控臭掩味;
陈化/黑蒜提取物(Aged Garlic / Black Garlic SAC Type)——鲜蒜控温湿发酵60~90天(28~35℃,RH 70%~90%) → 转化为S-烯丙基半胱氨酸(SAC) → 水/醇提 → 浓缩 → 喷雾干燥得深棕黑粉(SAC 0.3%~3% HPLC);
大蒜油(Garlic Oil)——超临界CO₂(35~40℃,15~25 MPa)或水蒸气蒸馏 → 油水分离 → 得淡黄色至橙红色挥发油(DADS/DATS为主)。
关键生产工序:
① 原料前处理(关键控制点)
选用山东(金乡/兰陵)、河南(杞县/中牟)、江苏(邳州)等产区3~4瓣白皮干蒜或鲜蒜,严控水分≤12%(干蒜)或无霉变(鲜蒜);入厂检测蒜氨酸(Alliin, HPLC)≥1.5%~2.0% DW,丙酮酸值(Allicin Yield)、农残、重金属(Pb≤5~10ppm);
干蒜/鲜蒜去皮、去芽(芽含苦味杂硫) → 依工艺选低温粉碎(≤15℃)或常温粉碎过10~20目筛;灭酶与保酶工艺严格分流(灭活线60~80℃短时,保酶线≤15℃避水)。
② 提取与酶解调控(Extraction & Enzymatic Control)
水/醇提取(Alliin/总硫):用 50%~70% 乙醇或去离子水,料液比1:5~1:10,控温≤50℃(N₂保护)浸提1~3h,防热敏降解与酶过度自解;合并滤液 → 减压浓缩(≤50℃);
酶解产Allicin(仅限稳定化/微囊/油线):破碎蒜泥在pH 5~7、20~37℃触发蒜氨酸酶反应15~60min(水相),生成Allicin后立即终止(升温≤50℃或酸调pH<4)或转入CO₂/溶剂萃取锁定;
超临界CO₂提油:蒜粉装釜,35~40℃、15~25 MPa萃取1~3h → 分离釜降压析油 → 得大蒜油(无溶剂残留)。
③ 纯化与分离(Purification & Separation)
蒜氨酸纯化:提取液经阳离子交换树脂(如732型)吸附 → 氨水解吸 → 低温浓缩 → 乙醇重结晶得Alliin ≥95%~98%白色结晶;
总硫/提取物富集:水/醇提液上大孔树脂(AB-8/D101) → 水洗 → 30%~70%乙醇洗脱 → 收集流份(HPLC跟踪Alliin/SAC/总硫);
膜分离(除杂浓缩):采用陶瓷膜(40~60nm)过滤除杂蛋白/胶体 → 纳滤(200~300Da)常温浓缩脱盐脱水,避免热敏损失;
微囊化(核心稳定技术):浓缩液加β-环糊精(β-CD)、N-LOK变性淀粉(8%~12%)低温均质/纳米射流雾化 → 喷雾干燥(进风150~170℃,出风70~80℃,壁材护活)得微囊粉,掩臭防氧化。
④ 陈化发酵(Aging / Black Garlic Process)
鲜蒜带皮入发酵罐,控温28~35℃、RH 70%~90%、60~90天,定期换气;期间Alliin/Allicin转化为SAC、黑色素、阿霍烯;发酵后去皮粉碎 → 水/醇提 → 浓缩 → 喷雾干燥。
⑤ 浓缩与干燥
非热敏线:喷雾干燥(进风150~170℃,出风70~80℃);
热敏/高纯线:真空/冷冻干燥(-40℃升华)防Allicin/Alliin降解;
大蒜油:低温真空蒸发除水/残溶。
⑥ 质控与包装
检测:HPLC测Alliin、SAC(OPA衍生荧光或UV@210~220nm)、有关硫化物;丙酮酸-DNPH法测Allicin Yield;UV测总硫/多酚;GC-MS测特征硫化物(DADS/DATS);重金属、微生物、稳定性(热/光/湿加速);
粉末:内双层棕铝箔袋+充氮;高纯Alliin/微囊避光、-20℃冷冻或2~8℃冷藏;大蒜油棕瓶充氮-20℃;保质期:微囊/灭活Alliin 24个月,活性酶粉12~18个月(防潮),黑蒜提取物18个月;

Process Route Overview:
Staherb produces five main lines of Garlic Extract/Allicin tailored to target stability/activity:
Enzyme-Inactivated Fixed Alliin Type (Human Oral/Pharma) — Fresh garlic low-temp milling → microwave/thermal enzyme inactivation (60–80℃ brief or microwave to prevent transient Allicin degradation) → low-temp water/EtOH extraction → cationic/macroporous resin purification → low-temp conc./freeze-drying → high Alliin (1%~98% HPLC) stable powder.
Active Enzyme-Retained Type (Food/Feed Alternative) — Fresh garlic milled ≤15℃ → no heating/pH adj → immediate freeze-drying (-40℃ sublimation) or ≤35℃ low-temp air drying → packaged; rehydration triggers Alliinase → Allicin generation in situ.
Stabilized Allicin / Microencapsulated Type — Enzymatic crude extract → β-cyclodextrin (β-CD) inclusion / N-LOK modified starch nano-jet atomization microencapsulation / enteric coating → low-temp drying to lock Allicin activity and control odor/masking.
Aged / Black Garlic Extract (SAC Type) — Fresh garlic fermented 60–90d (28–35℃, RH 70%–90%) → Alliin/Allicin converted to S-Allyl Cysteine (SAC), Ajoene, melanoidins → water/EtOH extraction → conc. → spray-dry to dark brown-black powder (SAC 0.3%–3% HPLC).
Garlic Oil — Supercritical CO₂ (35–40℃, 15–25 MPa) or steam distillation → oil-water separation → pale yellow to orange-red volatile oil (DADS/DATS dominant).

① Raw Material Pre-treatment
Source: Shandong (Jinxiang/Lanling), Henan (Qi/Zhongmu), Jiangsu (Pizhou) 3–4-clove white garlic (dry moisture ≤12% or fresh mold-free). Test: Alliin (HPLC) ≥1.5%~2.0% DW, pyruvic acid (Allicin Yield), pesticides, heavy metals (Pb≤5–10ppm); Peeled, germ-removed → low-temp milling (≤15℃) or ambient milling 10–20 mesh; enzyme inactivation vs retention strictly separated (inactivation 60–80℃ brief; retention ≤15℃ water-avoidance).
② Extraction & Enzymatic Control
Water/EtOH extraction (Alliin/Total Sulfur): 50%–70% EtOH or DI water, 1:5–10 w/v, ≤50℃ (N₂) soak 1–3h to prevent thermal/enzymatic degradation; combine → vacuum conc. (≤50℃).
Enzymatic Allicin generation (stabilized/oil only): Crushed garlic slurry at pH 5–7, 20–37℃ for 15–60min to trigger Alliinase → immediately terminate (↑≤50℃ or pH<4) or transfer to CO₂/solvent extraction locking.
Supercritical CO₂ Oil: Garlic powder in extractor, 35–40℃, 15–25 MPa, 1–3h → separate → collect garlic oil (solvent-free).
③ Purification & Separation
Alliin purification: Extract → cationic exchange resin (e.g., 732) adsorption → NH₄OH elution → low-temp conc. → EtOH recrystallization → Alliin ≥95%~98% white crystals.
Total Sulfur/Extract enrichment: Liquid on macroporous resin (AB-8/D101) → water wash → 30%–70% EtOH elution → collect fractions (HPLC-tracked Alliin/SAC/total S).
Membrane Separation: Ceramic membrane (40–60nm) filtration remove colloids/proteins → NF (200–300Da) conc. at ambient temp to avoid heat loss.
Microencapsulation (core stabilization): Conc. + β-CD, N-LOK modified starch (8%–12%) low-temp homogenization/nano-jet → spray-dry (inlet 150–170℃, outlet 70–80℃, wall-material protection) → micro-powder (odor-controlled, oxidation-resistant).
④ Aging / Black Garlic Fermentation
Fresh bulbs fermented 60–90d (28–35℃, RH 70%–90%) with ventilation; macromolecules degrade, Alliin/Allicin convert to SAC, melanoidins, Ajoene; post-ferment peel/mill → water/EtOH extract → conc. → spray-dry.
⑤ Concentration & Drying
Non-thermals: Spray-dry (inlet 150–170℃, outlet 70–80℃); Thermals/high-pure: vacuum/freeze-dry (-40℃ sublimation) to prevent Alliin/Allicin degradation; Garlic oil: low-temp vacuum evaporation for solvent/water removal.
⑥ QC & Packaging
Assay: HPLC Alliin, SAC (OPA-derivatized fluorescence or UV@210–220nm), related sulfurs; Pyruvic acid-DNPH method for Allicin Yield; UV total sulfur/polyphenols; GC-MS characteristic sulfides (DADS/DATS); heavy metals, microbes, stability (thermal/light/moisture acceleration).
Powder: inner brown foil + N₂-flush; high-pure Alliin/microcapsules light-protected, -20℃ or 2–8℃; Garlic oil amber bottle N₂ -20℃; shelf life: micro/inactivated Alliin 24 mos, active enzyme 12–18 mos (moisture-proof), AGE 18 mos.
Capacity: hundreds of tons/year extract, high-pure Alliin made-to-order mg to kg scale; Certifications: ISO 9001 / GMP / ISO 22000 / FSSC 22000 / HALAL / KOSHER; isolated Alliinase inactivation zone to prevent cross-activation.
大蒜素(Allicin)半衰期极短(体外数小时、体内<1分钟),严禁直接生产不稳定纯油用于人用口服;人用须酶灭活固定Alliin或微囊/肠溶/CO₂锁定;
蒜氨酸酶(Alliinase)遇热(>60℃长时)不可逆失活,灭酶与保酶工艺须严格分流;活性酶粉须绝对防潮(水触发酶解→Allicin瞬生即逝失效);
硫化物具强挥发性与刺激性,提取纯化全程N₂保护、避光、低温、密闭,防氧化降解与异味扩散;
陈化/黑蒜发酵须严控温湿周期防霉变与SAC峰值未达;
