Silodosin is an alpha-adrenergic receptor antagonist invented by Kissei Pharmaceutical Company in Japan, which has a very good effect on the treatment of urinary disorders associated with benign prostatic hyperplasia. The selective effect on urethra was 12 and 7.5 times higher than prazosin and Tamsulosin, respectively, and could significantly inhibit the prostate contraction induced by norepinephrine. It can inhibit bladder hyperactivity in a dose-dependent manner and increase the pressure threshold of bladder contraction in benign prostatic hypertrophy rats. These data suggest that cilodoxine, in addition to improving bladder function, is also effective in alleviating symptoms associated with benign prostatic hypertrophy. Compared to its peers Prazosin and Tamsulosin, silodoxin is highly selective towards α1A-receptors located in the prostate and bladder neck, while its affinity for α1B- and α1D- receptors is not significant. It blocks the alpha1A-receptor at this site, relaxes smooth muscle, and leads to improved urine flow and reduced BPH symptoms. Silodoxin binds more selectively to the α1A-receptor than to the cardiovasculary-associated α1b-receptor, maximizing activity to the target organ while minimizing potential effects on blood pressure.
