Protirelin is used clinically as a diagnostic agent to evaluate the function of the thyroid gland. It is administered as an intravenous injection and stimulates the release of thyroid-stimulating hormone (TSH) from the pituitary gland. The TSH then stimulates the thyroid gland to release the hormones thyroxine (T4) and triiodothyronine (T3).

Protirelin is also used in research to study the regulation of the hypothalamic-pituitary-thyroid axis and its relationship to various physiological processes.
