Linaclotide is a guanylate cyclase-C receptor agonist (GCCA) that has both visceral analgesic effects and endocrine active GC-C activation resulting in increased intracellular and extracellular cGMP concentrations. Extracellular cGMP can reduce the activity of painful nerve fibers and alleviate visceral pain in model animals. Intracellular cGMP can increase the secretion of chloride and bicarbonate in the small intestine by activating CFTR (cystic fibrosis transmembrane conduction regulatory factor), and ultimately lead to increased secretion of small intestinal fluid and rapid small intestinal transport.
Linaclotide is a peptide agonist of the guanylate cyclase 2C.
This compound reduces the activation of colonic sensory neurons, reducing pain; and activates colonic motor neurons, which increases smooth muscle contraction and thus promotes bowel movements.
