Chemical properties:
Imidazole is a five-membered aromatic heterocyclic compound containing two intermediate nitrogen atoms in the molecular structure. The unshared electron pairs of 1-position nitrogen atoms in the imidazole ring participate in ring conjugation, and the electron density of nitrogen atoms decreases, so that the hydrogen on this nitrogen atom is easy to leave in the form of hydrogen ions.
Therefore, imidazole is weakly acidic and can form salts with strong bases. The unshared electron pairs of the 3-position nitrogen atoms in the imidazole ring do not participate in conjugation, but occupy sp2 hybrid orbitals, can accept protons, and form salts with strong acids. Imidazole is slightly more basic than pyrazole and pyridine. There is tautomerism in imidazole ring.
Hydrogen at the 1-site nitrogen atom can be transferred to the 3-site nitrogen atom, so imidazole derivatives with the same substituents at the 4-site and the 5-site, respectively, are tautomers. Imidazole is stable to acid and has antioxidant properties. Imidazole derivatives exist widely in nature. Such as histamine, histidine, benzimidazole and so on.
Preparation method:
Using glyoxal as raw material, reacting with ammonium sulfate (or ammonia) in formaldehyde at 85 ~ 90℃, imidazole sulfate is prepared first, and then neutralized with calcium hydroxide, imidazole crude products can be obtained, filtered, washed with water, combined with filtrate and washing liquid, evaporation and concentration under reduced pressure, crystallization, can be prepared. If ammonia is used directly, a sulfate-free treatment step can be prepared in one step.
Whether ammonium sulfate or ammonia is used, the yield of this method is low, about 45%. Using o-phenylenediamine and formic acid as raw materials, cyclic synthesis, benzimidazole, and then oxidation in sulfuric acid solution, to generate dicarboxylic imidazole, and finally under the action of copper oxide, decarboxylated at 100 ~ 150℃, can produce crude products, and then recrystallized in benzene solution, imidazole product can be obtained.
Using D-tartaric acid as raw material, nitration with nitric acid in sulfuric acid to obtain 2, 3-dinitrotaric acid, and then react with ammonia in formaldehyde to obtain dicarboxylic imidazole, and then decarboxylated, can be prepared.
Use:
Determination of cobalt. Organic synthesis. Used for anti-metabolism, anti-histamine. Pharmaceutical pesticide intermediates, imidazole is an intermediate of the pesticide apromazole, imidazole and other fungicides, and is also an intermediate of the pharmaceutical antifungal drugs diclobenzimidazole, econazole, ketoconazole and clotrimazole. Used as curing agent for epoxy resin; Used as raw materials for imidazole derivatives, such as gambol, carbonyl diimidazole, etc. Effective buffer in the pH6.2-7.8 range; Used for titration of aspartate and glutamic acid.
