Growth hormone releasing peptide-6 can promote the secretion of growth hormone, regulate food intake, increase body weight, and promote animal growth
preparation
Peptide synthesis was performed under standard conditions using Rink resin in a self-shaking bed. In DMF, HBTU(3 equivalent) was used as the coupling reagent and DIEA(initially 3 equivalent, 1 equivalent 20 minutes after the coupling reaction began) was used as the base for amino acid coupling (3 equivalent). Fmoc is de-protected by treating the resin in DMF with 20% piperidine for 10 and 20 minutes. After each coupling and de-protection step, the resin is washed with DMF (2x), MeOH (2x), and DCM (2x), respectively. Side chain deprotection and Aza-Peptide cleavage. Freshly prepared TFA/H2O/TES solution (95:2.5:2.5,v/v/ v, 20 mL/g Aza-peptide resin) was then treated at room temperature for 2 h. The cleavage mixture is filtered and the resin is cleaned with a neat TFA. The filtrate was concentrated to about 1ml and treated with Et2O. The resulting azopeptide precipitates are filtered, washed with Et2O, dissolved in acetonitrile/water (1:1,v/v) solution, and lyophilized to produce a light foam or powder to obtain growth hormone releasing peptide-6.
