Use: Exemestane is a steroid aromatase inactivator, its structure is similar to the natural substrate of aromatase androstenedione, is the false substrate of aromatase
Synthesis: 6-methylene-androstene-4-ene-3, 17-dione (2) was synthesized from androstene-4-ene-3, 17-dione (3) by condensation with triethyl orthoformate, N-methylaniline and 40% formaldehyde solution, and then 1 was prepared by oxidation with 2, 3-dichloro-5,6 chemicalbook-dicyanoquinone (DDQ). Adding a certain amount of weak acid in oxidative dehydrogenation can shorten the reaction time from 15h to 8h, and the yield can be increased from 47% to 62%. The reaction conditions were mild, the separation was simple and the total yield was 45%.
