Indomethacin, an antiinflammatory agent, inhibits Cox-1 and Cox-2 (cyclooxygenases) with selectivity for Cox-1 (IC50 of 230 nM)over Cox-2 (IC50 of 630 nM). Additionally, indomethacin displays adipogenic actions via binding and activation of PPARγ(peroxisome proliferator-activated receptor γ), a ligand-activated transcription factor known to play an important role in adipogenesis. Other studies show indomethacin to stabilize cholesterol-dependent nanoclusters and enhance phase separation in biological membranes, which could consequentially effect plasma membrane assembled cell signaling cascades. Indomethacin is an activator of PPAR α.
Function
Indometacin has also been used clinically to delay premature labor, reduce amniotic fluid in polyhydramnios, and to close patent ductus arteriosus.
Indometacin is a potent drug with many serious side effects and should not be considered an analgesic for minor aches and pains or fever. The medication is better described as an anti-inflammatory, rather than an analgesic. Indometacin can also affect warfarin and subsequently raise INR.This product has obvious effect on anti-inflammation and antipyretic, can be easily absorbed and excreted. It can be fully absorbed after taken orally in the gastrointestinal tract, and in 1-4 hours the plasma concentration will reach a peak; it will be excreted by liver metabolism, with the urine.
