use
In mammals, β-nicotinamide mononucleotides are generated by Nicotinamide (Nam) catalyzed by Nampt (a protease in vivo), followed by NAD+ catalyzed by nicotinamide mononucleotide adenosyltransferase. Nicotinamide mononucleotide is a direct way to supplement NAD+. Niacinamide mononucleotides can reverse aging by significantly improving the repair ability of DNA damage in cells.
Nicotinamide mononucleotides are converted into "nicotinamide adenine dinucleotide (NAD)" which is essential for energy metabolism in the body. In mouse experiments, niacinamide mononucleotides have been shown to activate a gene called acetylase in the body, which can play a role in prolonging life and treating diabetes. NAD is a substance that the body can originally produce, and studies have shown that the amount of NAD in the body decreases with age.
preparation
Add 150ml of phosphate buffer solution to a 500ml four-mouth bottle.
Add 2g magnesium chloride, 10g ATP, 7g ribose, 10g niacinamide at a time, and stir at 35℃.
The pH value of the reaction system was detected, and the pH value was adjusted to 8.0 by 10% Na2CO3 solution. Then 1gRK enzyme powder and 2gNAMPT enzyme powder were added and dissolved by stirring.
The reaction was stirred at 200rpm at 37℃, and the conversion rate of the reaction was monitored by liquid chromatography-mass spectrometry. After 6 hours of reaction, ATP was detected to have been consumed and the reaction was stopped. After further filtration, the β-nicotinamide mononucleotide NMN was obtained by adsorption by HZ-818 macroporous resin, lyophilization, recrystallization with ethanol and water, and the yield was 70%.
