Glyburide (Glibenclamide) is a sulfonylurea compound that modulates insulin production. Sulfonylureas bind to ATP-dependent K+channels in beta cells of the pancreas, depolarizing them and stimulating the release of Ca2+, which in turnstimulates insulin production. Glyburide (Glibenclamide) is an ATP-dependent K+ channel (KIR6, KATP) and CFTR Cl- channel blocker.Thiscompound inhibits recombinant CFTR Cl- channels.
FunctionGlibenurea can inhibit glycogen decomposition and glycogen differentiation by increasing the insulin level of the portal vein or directly acting on the liver, and the hepatic generation and output glucose decrease. Oral absorption is fast, and the binding rate of protein is very high, which is 95%. After oral administration, the blood concentration reaches a peak of 2 ~ 5 hours and lasts for 24 hours. T1 over 2 is 10 hours. In liver metabolism, liver and kidney excretion each about 50%.
