WST-8 5-(2,4-disulfophenyl)-3-(2-methoxy-4-nitrophenyl)-2-(4-nitrophenyl)-2h-tetrazolium inner salt sodium salt (1:1) 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium,monoSodium sal (CAS:193149-74-5), in the presence of electronic carriers, is oxidized and reduced by dehydrogenase in cells to form water-soluble orange yellow methyl xanthate dye, which was soluble in tissue medium, and the amount of methylphen produced was proportional to the number of living cells. CCK-8 kit was prepared from WST-8 as raw material for the determination of the number of living cells in cell proliferation or toxicity experiments. It is a high sensitivity and non radioactive colorimetric method, which can replace the traditional MTT method. CCK-8 method is widely used in drug screening, cell proliferation, cytotoxic, tumor drug sensitivity, living cell count, cell activity and biological factor activity.
