1-Adamantanol, also known as 1-hydroxy amantadane, 1-tricyclic [3.3.1.1(3.7)] decanol, is a white crystalline powder at room temperature, melting point > 240℃, soluble in organic solvent, insoluble in water, has sublimation. Used in the manufacture of synthetic adamantane derivatives and Adabalin. Reaction with vinylidene chloride to obtain 1-amantadine acetic acid. 1-amantadine diacetic acid is then brominated, hydrolyzed to 3-hydroxy-1-amantadine acetic acid, and then reacted with vinylidene chloride to obtain 1, 3-amantadine diacetic acid. 1, 3-bis (4-acetylaminophenyl) amantadane can be directly synthesized by the Fouck alkylation reaction of 1-amantadanol and acetanilide in 98% sulfuric acid medium at room temperature, and 1, 3-bis (4-aniline) amantadane can be synthesized by alkaline hydrolysis. This synthesis route has the advantages of high reaction yield and easy purification. The synthesis of 1, 3-adamantane diaryl derivatives is greatly facilitated.
Application:
1-amantadanols are used as intermediates in the synthesis of amantadans. Used in the manufacture of synthetic adamantane derivatives and Adabalin. Reaction with vinylidene chloride to obtain 1-amantadine acetic acid.
