ピリジン誘導体
ピリジンの重要な誘導体はニコチン酸、コチン酸アミド、イソニアジド、ニコチン、ブルシン、ビタミンB6等を有する。 ビタミンB6は補酵素作用を有する四置換ピリジン誘導体の一種である。臨床で妊娠嘔吐、放射性嘔吐、イソニアジド中毒症、脂漏性皮膚炎とナイアシン欠乏症等の治療に用いられる。 ピリジンは一種の弱アルカリ性と特殊な臭いを有する無色又はわずかに黄色の液体であり,コールタール、シェール油、石炭ガス及び石油中に天然に存在し,水、アルコール、エーテル、石油エーテル、油類、ベンゼン、クロロホルム等の多くの有機溶媒と融合でき,重要な化学原料と溶媒である。 ピリジンは略正六角形の構造を有し,ベンゼンと類似し,同じ電子構造を有する。環中の窒素原子が電子吸引性を有するため,2,4,6位における電子雲の密度は3,5位より低く,酸性媒質中で,求電子置換反応は3,5位において行い,それに対して求核反応例えばアミン化、アルキル化、アリール化、アシル化は2,4,6位において行う。ピリジンは一種の弱い第三級アミンであり,エタノール溶液で複数の酸(例えばピクリン酸、過塩素酸等)とともに水に溶けない塩を形成することができる;ピリジンはアルカリ性を呈するため,塩酸と反応してピリジン塩酸塩(C5H5N•HCl)を生成することができる。ニッケル触媒の作用で,200℃と15~30MPaの環境下で,水素還元反応を行い,ピペリジンを生成することができる;また電解還元によりピペリジンを生成することもできる;ピリジンの還元性はベンゼンより高い。ピリジンの酸化性はベンゼンより低いが,過酸化水素又は過酢酸を利用してピリジンを酸化させてN-オキシドピリジンを生成でき,これは一種の重要なピリジン誘導体であり,窒素原子は酸化された後に,正電荷を有するピリジンイオンを形成できないため,アリール基の求電子置換反応に役立つ。ピリジンの求電子置換例えばニトロ化、スルホン化、ハロゲン化はいずれも難しいが,そのうちハロゲン化はその他の二つより簡単であり,200℃以上の温度で,3,5-ジクロロピリジン、又は3,4.5-トリクロサンピリジンを取得できる。ピリジンは複数の金属イオンとともに結晶性 配位化合物を形成することができる。
