The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Glypican 1 (GPC1) is located at 2q37.3 on the human chromosome. It belongs to the family of glycosylphosphatidylinositol-anchored heparan sulfate proteoglycan. GPC1 is localized in the vascular system. GPC1 consists of an N-terminal secretory signal peptide, 14 conserved cysteine residues, a heparan sulfate (HS) attachment domain near the C terminus and a hydrophobic domain for attachment of the glycosylphosphatidylinositol (GPI) at the C terminus.
Biochem/physiol Actions
Glypican 1 (GPC1) plays an important role in cell growth, differentiation and morphogenesis. It also plays a role in cell migration, adhesion, anti-coagulation, lipoprotein metabolism and modulation of growth factors. GPC1 exhibits chaperone-like activity by restoring the binding activity of oxidized VEGF165 (vascular endothelial growth factors-165). Overexpression of GPC1 results in over accumulation of β-amyloid protein (Aβ) in the Alzheimer′s disease (AD) brain. Mutation in this gene is associated with Niemann-Pick C1 disease.
