RO-52 (SSA), also called as tripartite motif-containing protein 21 (TRIM21) is a 52 kDa, ubiquitin E3 ligase. It has a N-terminal RING finger domain (Really Interesting New Gene), crucial for ubiquitination functionality. RO-52 (SSA) gene is mapped to human chromosome 11p5.5. It is capable of autoubiquitination and uniquitylates p62 protein and regulates redox homeostasis. Patients with Sjogren′s syndrome display elevated levels of anti-Ro/SSA antibodies. RO-52 levels in peripheral blood mononuclear cells (PBMC) contributes to pathogenesis of systemic lupus erythematosus (SLE), systemic autoimmune rheumatic diseases (SARDs), interstitial lung disease (ILD) and connective tissue diseases (CTD). RO-52 is considered as potential target for treating SLE.
