Uses
5-Chloro-N-methylthiophene-2-carboxamide is an impurity of Rivaroxaban (R538000), which is a novel antithrombotic agent. A highly potent and selective, direct FXa inhibitor.
Synthesis
Step add 5-chlorothiophene-2-carboxylic acid, dichloromethane, dichlorosulfoxide dropwise at 0 , dropwise addition, room temperature stirring for 4 hours;
Step dropwise addition of aqueous solution with a mass concentration of 40% of methylamine at 0 , dropwise addition is completed, stirring at room temperature for 18 hours;
Step after the extraction of filtration, the cake is dried at 55 , the solid is 5-chlorothiophene-2-carboxymethylamine;
5-chlorothiophene-2-carboxylic acid, dichloromethane, dichlorosulfoxide, 40% methylamine. The solid is 5-chlorothiophene-2-carboxamide;
