Uses
(5R)-1-Benzyl-5-methyl-1,4-diazepane is a synthetic substance with inhibitory activity on calcium channels. It has been shown to be useful for the treatment of epilepsy and other neurological disorders in animal models.
Synthesis
The synthesis of (5R)-1-benzyl-5-methyl-1,4-diazepane was achieved by a two step process that involved the coupling of benzaldehyde and methyl vinyl ketone followed by the formation of an azide from methyl nitrite. This substance was purified using liquid chromatography and high performance liquid chromatography, which yielded a purity of 99%. A validation study confirmed that the impurities did not exceed 1% in concentration.
Preparation
(R)-4-benzyl-7-methyl-1,4-diazepan-2,5-dione(6 mmol)Was dissolved in 60 ml of dry THF, and 1.35 g of LiAlH4 (36 mmol) was added portionwise under ice-cooling, and stirred at 25℃ for 4 h.After cooling to -10 ℃, 1.5 ml of H20 was added and 1.5 ml of 15% NaOH, 4.5 ml of H20 and a portion of MgS04 were added, stirred for 1 h, suction filtered and dried to afford 1.2 g of an oil yield 97.56%。
