- for the anti-mouse IgG conjugated alkaline phosphatase in immunoblotting
- for alkaline phosphatase in the histochemical staining of primary osteoblasts
- for alkaline phosphatase staining in bone marrow stromal cells
Uses
A ready-to-use, precipitating substrate system for alkaline phosphatase. This substrate system produces an insoluble NBT diformazan end product that is blue to purple in color and can be observed visually. Immunohistochemical staining with BCIP/NBT utilizes requires organic mounting media and can be counterstained with Nuclear Fast Red or Light Green. BCIP/NBT is generally a more sensitive method than Fast Red. Not recommended for ELISA (microwell) procedures.
General Description
BM purple is a chromogenic substrate for alkaline phosphatase (AP) designed for precipitating enzyme immunoassays. It develops a permanent, dark purple band or spot at the AP binding site on the membrane or solid support. The stabilized substrate solution may be used directly from the bottle with no mixing or reconstitution. It gives less background staining than when using the BCIP/NBT two-bottle system, without a loss of sensitivity. The reaction product has a dark purple color and is insoluble in water. The dark purple color can be visually seen.
