Lomibuvir is a non-nucleoside inhibitor of the RNA-directed RNA polymerase of hepatitis C virus (HCV; K
i = 17 nM) that inhibits primer-dependent RNA synthesis selectively over
de novo RNA synthesis. It inhibits HCV replication in Huh7.5 cells expressing the 1b/Con1 HCV subgenomic replicon (EC
50 = 5 nM).
