General Description
4′-Chloroacetanilide is the main impurity present in acetaminophen and has been quantitated by new high-performance liquid chromatography method using a mixed-mode reversed-phase/cation exchange stationary phase.
Synthesis
Preparation of p-chloroacetanilide: 113.5 mg (0.5 mmol) of 4-chlorophenyl diazonium tetrafluoroborate, 0.1 mmol of cuprous iodide, 0.5 mmol of potassium phosphate with 1.0 mmol of water were added to a Schlenk tube under nitrogen and 1 mL of acetonitrile was added under air and the reaction was stirred for 12 hr at 80oC, the yield of the reaction was 68%.
Purification Methods
Crystallise the anilide from EtOH or aqueous EtOH. [Beilstein 12 IV 1178.]
