BV 6性质、用途与生产工艺

生物活性 BV-6是一种SMAC模拟的cIAP和XIAP的双重抑制剂。
体外研究 BV6抑制HCC193 NSCLC细胞的细胞活性，IC50 为 7.2 μM，在HCC193 和H460细胞系引起细胞凋亡，并通过分别激活裂解的caspase-8 和caspase-9显著增加这些细胞系的放射敏感性。在不成熟的树突细胞中，BV-6治疗导致经典的NF-kB通路适度激活。此外，BV-6增加CIK细胞介导的血液病(H9，THP-1，和Tanoue)和固体肿瘤(RH1，RH30，和TE671)的消退。BV-6也会增强外周血单核细胞的凋亡，并且对免疫细胞限制其细胞毒性潜能具有显著的抑制作用。

体内研究

Murine cIAP-1, cIAP-2 and XIAP expressions are clearly observed in the cytoplasm of both epithelial and stromal cells of implants, whereas Survivin is mainly expressed in the nuclei BV6 treatment for 4 weeks attenuated the intensity of IAPs expression. The size of lesions range from ~2 to 7 mm in diameter. The monolayer epithelial cell lining of the cyst is shown. After immunohistochemical staining, cytokeratin and vimentin are positively stained, whereas calretinin is negative. After BV6 treatment for 4 weeks, the total number of lesions (4.6 versus 2.8/mouse), the average weight (78.1 versus 32.0 mg/mouse) and the surface area (44.5 versus 24.6 mm ² /mouse) of lesions are significantly less than in the controls. In the endometrial gland epithelia or stroma, the percentage of Ki67-positive cells decreases after BV6 treatment.
