  • 背景 Anti-Human secondary antibodies are affinity-purified antibodies with well-characterized specificity for human immunoglobulins and are useful in the detection, sorting or purification of its specified target. Secondary antibodies offer increased versatility enabling users to use many detection systems (e.g. HRP, AP, fluorescence). They can also provide greater sensitivity through signal amplification as multiple secondary antibodies can bind to a single primary antibody. Most commonly, secondary antibodies are generated by immunizing the host animal with a pooled population of immunoglobulins from the target species and can be further purified and modified (i.e. immunoaffinity chromatography, antibody fragmentation, label conjugation, etc.) to generate highly specific reagents.
  • 检测方法 正常情况下血清IgE仅在ng/ml水平，用常规测定IgG或IgM的凝胶扩散法检测不出IgE，必须用高度敏感的放射免疫测定法及酶联免疫测定法进行检测。
    1．放射免疫吸附剂试验（radioimmunosorbenttest，IRST）是将抗IgE吸附到固相载体上用以检测血清IgE的方法，故又称固相放射免疫测定（SPRIA）；临床常用双抗体夹心法，多以滤纸为载体。将抗IgE抗体偶联到经溴化氟活化的滤纸上，使其与待检血清及IgE参考标准进行反应；洗涤后加入125I标记的抗人IgE，再经洗涤后测定滤纸片的放射活性，其测定值与标本中的IgE含量呈正相关。
    2．酶联免疫测定法测定血清IgE时也常用双抗体夹心ELISA法，操作方便，敏感性也很高，在临床上经常应用。
    3．间接血凝试验用抗IgE至敏红细胞，将标本血清做系列稀释后与致敏红细胞反应。此法更加简便易行，便于普及，但敏感性比上两法稍低。
