The ubiquitin C-terminal hydrolase L1 (UCH-L1) gene is mapped to human chromosome 4p13. The protein localized in the cytosol and nucleus. UCH-L1 protein is primarily expressed in neuroendocrine tissues, neurons and in testis/ovary. Ubiquitin C-terminal hydrolase-L1 (UCHL1) is a deubiquitinating enzyme (DUB) that hydrolyzes C-terminal ubiquitin esters and amides, and peptide?ubiquitin amides to ubiquitin monomers in- vitro. In addition to DUB activity, it also possesses E3 ubiquitin-protein ligase activity. UCHL1 is involved in the regulation of the ubiquitin-dependent proteolytic system. UCH-L1 plays a vital role in cancer development and progression by upstream activation of protein kinase B (Akt). Mutation in the UCHL1 gene is associated with the development of Parkinson′s disease (PD).
