Immunocytochemistry/ Immunofluorescence - Anti-Tyrosine Hydroxylase antibody (ab137869)
Immunocytochemistry/ Immunofluorescence analysis of C6 (Rat glial tumor cell line) cells labeling Tyrosine Hydroxylase with Purified ab137869 at 1:100 dilution (5.6μg/ml). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1:200 (2.5 μg/ml). ab150077 Goat anti rabbit IgG(Alexa Fluor® 488) was used as the secondary antibody at 1:1000 dilution. DAPI nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.
酪氨酸羟化酶(Tyrosine hydroxylase)是负责催化氨基酸L-酪氨酸转变为二羟基苯丙氨酸(多巴)的酶。因此它使用四氢生物蝶呤作为辅酶。多巴是多巴胺的一个前体,相应地,后者亦是去甲肾上腺素与肾上腺素的前体。在人体中,酪氨酸羟化酶由TH基因编码出来。
酪氨酸羟化酶可以被药物α-甲基-对-酪氨酸(甲基酪氨酸)所抑制。此抑制作用可以导致脑部多巴胺与去甲肾上腺素的消耗,这是因为缺乏前体L-多巴(L-3,4-二羟基苯丙氨酸),此物质可以由酪氨酸羟化酶所合成。此药很少被使用并会导致抑郁,但它在治疗嗜铬细胞瘤以及抗高血压方面很有用处。
双抗体夹心法测定标本中Tyrosine Hydroxylase水平。用纯化的Tyrosine Hydroxylase抗体包被微孔板,制成固相抗体,往包被单抗的微孔中依次加入Tyrosine Hydroxylase,再与HRP标记的Tyrosine Hydroxylase抗体结合,形成抗体-抗原-酶标抗体复合物,经过彻底洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色,并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的Tyrosine Hydroxylase呈正相关。用酶标仪在450nm波长下测定吸光度(OD值),通过标准曲线计算样品中Tyrosine Hydroxylase浓度。
