以2-吡啶甲醛肟(288a,180 mg,1.47 mmol)为原料,在20℃下,将其溶于DMF(2 mL)中。随后,一次性加入N-氯代丁二酰亚胺(NCS,216 mg,1.62 mmol)。反应混合物在20℃下搅拌12小时。反应完成后,将混合物浓缩。将残余物倒入水(20 mL)中,用乙酸乙酯(50 mL × 3)萃取水相。合并的有机相用盐水(20 mL × 2)洗涤,无水Na2SO4干燥,过滤并真空浓缩,得到2-吡啶氯化甲醛肟(288b,200 mg,收率:87.1%),为棕色固体。1H NMR(400 MHz,CDCl3)δ 10.79(br s,1H),8.72(d,J = 4.2 Hz,1H),7.94(d,J = 8.2 Hz,1H),7.80(dt,J = 1.7, 7.8 Hz,1H),7.39(ddd,J = 0.8, 5.0, 7.4 Hz,1H),2.79(s,1H)。
